Tombstone

Developed as a dark counterpart to Spider-Man, Venom in the comics first appeared with Eddie Brock as its host. But in a surprising twist, Spider-Man 2 uses Peter Parker's childhood friend Harry Osborn instead as the symbiote's host.
The Scorpion

The Scorpion's origin in the game is drastically different from his private investigator counterpart in the comics. Instead, he is a fiercer, more intimidating villain in Spider-Man 2 while still keeping the scorpion tail and suit. Though the player won't be fighting him, he has a few memorable cutscenes.

Spider-Man 2's Scream has a unique twist on the character, who is normally a Venom villain. As one of Venom's “children”, the red and yellow symbiote possesses Mary Jane Watson, forcing Peter Parker to face her in a boss battle where he eventually frees her.
Symbiote Spider-Man/Black-Suit Peter

First as a playable character but later on as an eventual boss fight, Symbiote Spider-Man will be battled during the “Anything Can Be Broken” Side Mission with the player controlling Miles Morales.
Despite giving Peter Parker enhanced strength and other abilities, the Symbiote Spider-Man suit eventually turns violent by tapping into Peter Parker's darker impulses, forcing Miles Morales to battle him.
Doctor Octopus

Kraven the Hunter

Preferring to hunt with his bare hands, Kraven will be a worthy challenge for players who want a visceral Spider-Man experience.
