To get to the Hospital Wing, go through the locked door in the Faculty Tower. After unlocking the door with Alohomora and passing through, go up the stairs straight ahead to reach the Hospital Wing on the top floor. You'll know you've entered the Hospital Wing as there are beds lined up.

The door leading to the Hospital Wing will not open unless you play the main quest "The Caretaker's Lunar Lament". If the quest hasn't appeared yet, progress through the main story first and play the quest.

When you can't go to the Hospital Wing, progress the story and learn the incantation for Alohomora. You can obtain the Alohomora spell by clearing the main quest "The Caretaker's Lunar Lament".
No Need to Enhance Alohomora
If you only need to go to the Hospital Wing, there is no need to enhance Alohomora. Alohomora can be enhanced by clearing the side quest "The Man Behind the Moons", allowing you to unlock doors of higher levels. However, the door leading to the Hospital Wing is level 1, so it is unnecessary.
